Abstract
In this study, small intestine submucosa (SIS) - hyaluronic acid (HA) sponges were prepared by freeze-drying SIS-HA mixtures and characterized for the possible application of tissue engineered scaffold. HA of ratios of 1, 2, 5 or 10% was added to SIS dissolved in 3% (v/v) acetic acid solution. The prepared sponges were crosslinked by 50 mM of EDC solution and were lyophilized. To analyze chemical and physical properties of the scaffold, SEM, XRD, FTIR, compression test and water uptake test were used. WST assay and H&E stain was used to measure the proliferation and infiltration of adipose derived stem cell, respectively. ALP assay was used to evaluate the expression of osteo-differentiation. Our study revealed that HA ratio affected cell viability and bone differentiation. In conclusion, the composite scaffold of SIS-HA5% sponge could be considered a potential bone tissue engineering scaffold.

본 연구에서는 소장점막하조직(SIS)에 히알루론산(HA) 함량을 달리하여 동결건조법으로 SIS-HA 스폰지를 제작하였고 그 특성을 분석함으로써 생체조직공학적 지지체로서의 응용 가능성을 확인하였다. 3% 아세트산 용액에 SIS와 SIS 함량의 1, 2, 5 및 10% HA를 첨가하여 스폰지를 제작하였고 50 mM의 가교제를 통해 가교하여 이를 전자주사현미경, X-ray 회절분석, 적외선 분광기, 기계적 강도 및 물 흡수성 실험을 통해서 물리, 화학적 분석을 시행하였다. 지지체 내에서 지방세포유래 중간엽줄기세포의 증식률과 침투성은 WST 분석과 H&E 염색으로 확인하였고 초기의 골분화능은 ALP 분석으로 확인하였다. 이러한 결과를 통하여 세포 성장률 및 골분화능에 HA 함량이 영향을 미치는 것으로 판단되었고 제조된 SIS-HA5% 스폰지는 골재생에 있어서 조직공학적 지지체로서의 가능성을 보여주었다.
