Abstract
In this study, we present preparation of the gellan gum (GEL)-croaker swim bladder (CSB) membrane and evaluation of its use as a bone regeneration membrane. GEL/CSB membranes were obtained by casting and drying the GEL/CSB solution and adding D-PBS (Mg2+, Ca2+, K+). The effects of composition and crosslinking on the physicochemical properties of GEL/CSB membranes were evaluated by FTIR analysis, XRD, swelling ratio, enzymatic degradability and cell viability test. Also, MC3T3-E1, pre-osteoblast cell line, were treated with GEL/CSB of various concentrations (2.5-100 μg/mL). And then, the effect of GEL/CSB on osteoblast differentiation was examined by alkaline phosphatase (ALP) activity. ALP activity increased as the applied GEL/CSB concentrations increased in a dose-dependent manner. The bone regeneration was evaluated by micro-computed tomography (micro-CT) analysis at 4 weeks postoperatively in rat. The GEL/CSB membrane has significant effect on the new bone formation and has the potential to be applied for guided bone regeneration.

본 연구는 골 재생 막으로 사용하기 위해 젤란검(GEL)/민어부레추출물(CSB)을 막으로 제조하고 평가하였다. GEL과 CSB 용액을 캐스팅하고 건조시킨 후 D-PBS(Mg2+, Ca2+, K+)을 첨가하여 GEL/CSB 조성 막을 제조하였다. GEL/CSB 막의 물리-화학적인 특성을 분석하기 위해 FTIR, XRD, 팽윤도, 효소 분해도, 세포 생존율을 평가하였다. 또한 조골세포 MC3T3-E1에 GEL/CSB 조성 막을 다양한 농도로 처리하여 alkaline phosphatase(ALP) 활성을 측정하였다. GEL/CSB 농도가 증가함에 따라 ALP가 증가하였다. 막을 처리한 rat의 골 재생률을 수술 4주 후, 마이크로 CT를 이용하여 관찰하였다. GEL/CSB 막이 새로운 신생골 형성에 상당한 효과가 있는 것을 확인하였다.
